چکیده

Four hundred isolates obtained from soil samples collected from forest, river basin, agricultural fields and rhizosphere of plantation crops of North Bengal were screened for phosphate solubilizing activity on Pikovskaya’s agar medium. Among the screened isolates, ninety showed phosphate solubilizing activity. Out of these, ten isolates belonging to Aspergillus niger, A. melleus and A. clavatus were selected for further studies. In vitro evaluation of phosphate solubilization by the different isolates using tricalcium phosphate (TCP) and rock phosphate (RP) revealed that the isolates could solubilize TCP better than RP. Selected isolates were mass multiplied using farm-yard manure (FYM) and were tested in vivo for their growth promoting activity in soybean. While all the isolates promoted growth, A. niger RSP-14 was found to be most effective. While the soil P content decreased due to the activity of the PSFs, root phosphate content showed an increase. These isolates were further analyzed for genetic variability. Genomic DNA from the fungal isolates were obtained and purified. UPGMA cluster analysis divided the ten isolates into two groups with the genetic similarity ranging from 0.350.61. One group consisted of four isolates of A. niger and five isolates of A. melleus, while the other group had one isolate of A. clavatus.

Molecular Characterization of Trichoderma viride and Trichoderma harzianum Isolated from Soils of North Bengal Based on rDNA Markers and Analysis of Their PCR-RAPD Profiles

Nineteen isolates of Trichoderma viride and Trichoderma harzianum obtained from rhizosphere soil of plantation crops, forest soil and agricultural fields of North Bengal region were studied using RAPD and ITS-PCR. The genetic relatedness among eleven isolates of T. viride and eight isolates of T. harzianum were analyzed with six random primers. RAPD profiles showed genetic diversity among the i...

Diversity Status of Beneficial Microflora in Saline Soils of Tamil Nadu and Pudhucherry in Southern India

Alleviating plant salt stress and remediating saline soils are of great economic interest. Beneficial microbes such as Arbuscular Mycorrhizal (AM) fungi, Plant Growth Promoting Rhizobacteria (PGPRs), etc. are associated with many plants including trees. These beneficial microbes can cope up with salinity and help the plants to survive in such soils. In this study, diversity status of beneficial...

Species diversity of indigenous Trichoderma from alkaline pistachio soils in Iran

The diversity of Trichoderma spp. was investigated in alkaline soils of pistachio orchards at different geographic areas in Kerman province, Iran. A total of 161 Trichoderma isolates were obtained and identified at the species level by their morphological characters and sequence analysis of their internal transcribed spacer (ITS) and tef1-α genomic regions. Totally, five species of Trichoderma ...

Characterization of zinc-solubilizing Bacillus isolates and their potential to influence zinc assimilation in soybean seeds.

One hundred thirty-four putative Bacillus isolates were recovered from soybean rhizosphere soils of Nimar region to select effective zinc solubilizers for increased assimilation of zinc (Zn) in soybean seeds. These isolates were screened in vitro for zinc-solubilization ability on Tris-minimal agar medium supplemented separately with 0.1% zinc in the form of zinc oxide, zinc phosphate, and zinc...
